Structural characterization of the latent complex between transforming growth factor β1 and β1-latency-associated peptide

چکیده
The formation of a non-covalent complex between mature transforming growth factor β1 (TGF-β1) and its pro region, the β1-latency-associated peptide (β1-LAP), is important in regulating the activity of this multipotent growth factor. We have overexpressed simian β1-LAP in Chinese hamster ovary (CHO) cells to produce a cell line which secretes β1-LAP into the culture medium at " 1 mg}l, thus enabling structural studies of complex formation between β1-LAP and TGF-β1. The simian β1-LAP expressed in CHO cells reversed the growth inhibitory effect of exogenous TGF-β1 on Mv1Lu (mink lung epithelial) cells and was able to form a cross-linked complex with "#&I-TGF-β1. Simian β1-LAP was purified to homogeneity by a combination

Human Leukocyte Antigen-G Expression on Dendritic Cells Induced by Transforming Growth Factor-β1 and CD4+ T Cells Proliferation
Background: During antigen capture and processing, mature dendritic cells (DC) express large amounts of peptide-MHC complexes and accessory molecules on their surface. DC are antigen-presenting cells that have an important role in tolerance and autoimmunity. The transforming growth factor-beta1 (TGF-β1) cytokine has a regulatory role on the immune and non-immune cells. The aim of this study is ...
متن کاملThe Effect of Regular Swimming Exercise on the Levels of Renal Matrix Mettaloproteinase-2 and Transforming Growth Factor-β1 in Rats with Diabetes
Background & Aims: Numerous studies have reported the renoprotective effects of exercise in both human and animal models of diabetic nephropathy. However, detailed mechanism of action by which exercise has a favorable influence on renal fibrogenic factors is not yet fully understood. Therefore, the aim of this study was to assess the effect of swimming exercise on the activity ...
متن کاملIncreased angiogenesis in primary myelofibrosis: latent transforming growth factor-β as a possible angiogenic factor
OBJECTIVE The aim of this work was to demonstrate a possible relationship between anti-latency-associated peptide human latent transforming growth factor beta 1 (latent TGF-β1) expression in megakaryocytes and microvascular density in bone marrow biopsies from patients with essential thrombocythemia and primary myelofibrosis. METHODS Microvascular density was evaluated by immunohistochemical ...
متن کاملCo‐expression of the protease furin in Nicotiana benthamiana leads to efficient processing of latent transforming growth factor‐β1 into a biologically active protein
Transforming growth factor beta (TGF-β) is a signalling molecule that plays a key role in developmental and immunological processes in mammals. Three TGF-β isoforms exist in humans, and each isoform has unique therapeutic potential. Plants offer a platform for the production of recombinant proteins, which is cheap and easy to scale up and has a low risk of contamination with human pathogens. TG...
متن کاملMaternal Serum Levels of Transforming Growth Factor ß1 (TGF-ß1) in Normal and Preeclamptic Pregnancies
Background: Successful pregnancy in allopregnant women depends upon the control of graft rejection mechanisms. It has been suggested that some immunosuppressive cytokines contribute to successful pregnancy and transplantation. Transforming growth factor beta (TGF- β) exhibits potent immunoregulatory and anti-inflammatory properties which might prolong graft survival. Recent studies suggest a ...
